TCRb repertoire of DN and large DP thymocytes from mice expressing a wild type or mutant CD3zeta chain

Mice expressing a CD3zeta chain with a mutation in the transmembrane domain show impaired beta-selection, coinciding with reduced pre-TCR signaling capacity. To study changes in the early TCR repertoire, DN and large DP thymocytes were purified form CD3zeta wild type and mutant mice and TRBV-encoding mRNAs were sequenced after unbiased PCR expansion.
