We employed three approaches to evaluate the roles of membrane lipids in salt stress responses. First, we used the fluorescent dye propidium iodide (PI) at a concentration of 50 μg mL⁻¹ to assess membrane integrity. Five isolated strains were cultured in potato dextrose broth (PDB) for 1 week, and their hyphae were stained with PI. Next, we utilized Nile Red at a concentration of 2.5 μg mL⁻¹ to visualize the hyphal lipid droplets that protect against membrane lipid peroxidation under diverse stress conditions. The size of the lipid droplets was measured using ImageJ software (National Institutes of Health, Bethesda, MD, USA). Third, we estimated salt-induced membrane lipid peroxidation by quantifying the MDA content at NaCl concentrations of 0, 0.3 M, and 0.8 M.
