RNA-Seq analysis was carried out to investigate the effects of selective CDK8/19 inhibitor Senexin B (different treatment period) and knockout/re-expression of CDK8 and CDK19 proteins) on gene transcription in HEK293 cells Overall design: Parental HEK293 cells (WT), CDK8/CDK19 single-knockout derivatives (8KO, 19KO), CDK8/19 double-knockout (dKO), and dKO-reexpression derivatives (dKO-V, dKO-8, dKO-19) were treated with Vehicle control (XXX.C) or 1µM Senexin B (XXX.S) for different period of time (3hr, 5hr, 24hr, 3d and 15d). Each treatment condition was done in biological triplicate.
