Using bulk RNA-seq, the transcriptional profile of two cell lines with high or low metastatic properties derived from single clones of the murine 4T1 cell line treated or untreated with iron chelator DFX was analyzed in order to determine the mechanism underlying the inhibition preference of DFX for highly metastatic 4T1 cells. Overall design: 1) Single clonal lines were established using single limiting dilution from the 4T1 mammary cancer cell line. 2) Using in vitro and in vivo study to determine the metastatic property we selected 2 single clones to represent highly metastatic(HM) and lowly metastatic(LM) 4T1 cell lines. 3) 2 single clones were treated with 50 uM DFX or equivalent dose of DMSO as treatment and control groups for 48-hour. 4) Total RNA was extract and carried out using bulk RNA-seq.
