Long non-coding RNAs (lncRNAs) have been found to exhibit strong regulatory functions in cell physiology and maturation of dendritic cells (DCs), which are one of the critical immune cells in systemic lupus erythematosus (SLE). Hence, this study tried to reveal the underlying roles of one lncRNA, lncTAF15:1-1, in modulating DCs functions and its involvement in SLE pathogenesis.Monocyte-derived dendritic cells (moDCs) were cultivated and differentiated in vitro. Expression levels of lncTAF15:1-1 were compared using qPCR. Comparisons of moDCs biology functions including cell surface makers, secretion levels of inflammatory cytokines, and migration ability were performed by flow cytometry, ELISA, and Transwell chamber experiment. RNA-seq of lncTAF15:1-1 overexpression cells and negative control cells were then conducted to detect molecular alterations at the transcriptional level. Gene Set Enrichment Analysis (GSEA) analyses were utilized to predict potential involved signaling pathways, which were then confirmed by Western Blotting.
