Study on the characteristics of the immune microenvironment and skin microbiome in eczema based on multi-omics analysis

Three eczema skin samples and two control skin samples were randomly selected. The samples were digested with Collagenase IV and DNase I at 37 for 1 hour, followed by Ficoll density gradient centrifugation to remove dead cells and debris. Single-cell capture and mRNA amplification were performed using the 10x Genomics Chromium single-cell platform, and cDNA libraries were constructed using the Chromium Single Cell 3 v3 Kit. Library quality control was conducted using the Agilent 2100 Bioanalyzer and qPCR to assess concentration and fragment distribution. The libraries were then sequenced on the Illumina NovaSeq 6000 platform.
