A plane wave generated by a He-Ne laser with a 632.8 nm wavelength (Newport N-STP-912) is collimated and expanded. The beam then passes through a beam splitter (BS), modulated and reflected by a phase-only spatial light modulator (SLM) (Holoeye LETO-3-VIS-009), and reflected again by the BS before being transmitted to a 4f system consisting of two Fourier lenses with an 80mm focal length. A sCMOS detector (PCO edge4.2) is positioned ~5mm behind the rear focal plane of the 4f system to capture the diffracted patterns. We use non-sparse dynamic microorganisms as our phase objects.
