RNAsequencing comparing WT S. aureus and an evolved mutant EVOL-A and EVOL-A ?sarA

The goals of this experiment were to understand the basal changes in transcription between the EVOL-A strain that is an evolved strain in the NRS193 background with a mutation in Rel (Q672*) that has unknown impacts on transcription. The EVOL-A delSarA mutant was used to understand which changes in EVOL-A comapred to WT (NRS193) are dependent on the DNA-binding protein SarA. Overall design: S. aurues strains WT NRS193 (WT), EVOL-A (EV), and EVOL-A ?sarA (delsarA) were grown overnight in TSB at 37C shaking and then back diluted 1:10 in fresh TSB for 1 hour. From this subculture RNA was prepared for sequencing. Performed in three biological replicates.
