Transcriptomic Analysis of adah-1 and pnp-1 RNAi treated C. elegans

RNA-seq was used to identify gene expression changes in C. elegans upon adah-1 and pnp-1 RNAi treatment Overall design: Ten C. elegans N2 at L4 stage (P0) were picked onto RNAi plates seeded with adah-1, pnp-1, or L4440 cultures in E. coli HT115 background and incubated at 20°C until their progeny (F1) became gravid adults, which were bleached to obtain synchronized L1 progeny. 1500 L1 progeny (F2) were plated on two 10cm RNAi plates seeded with the respective RNAi culture. These animals were incubated at 20°C for 56 hours, at which point animals were adults and collected for RNA extraction.
