This project entails the systematic characterization of poly(A) site choice in Arabidopsis leaves and seeds. Custom-made cDNA tags that query the mRNA-poly(A) junction were prepared and sequenced on the Illumina GAII platform. Thee tags were then curated and mapped to the Arabidopsis genome. From these studies, a global overview of poly(A) site choice has been obtained, and numerous instances of alternative polyadenylation and antisense transcription identified.
