An orthotopic syngeneic mouse model of bortezomib-resistant multiple myeloma

RNA was extracted from myeloma cell lines which were made engraftable and resistant to bortezomib and the transcriptome was characterised using RNA sequencing. Overall design: 5TGM1. wt cells were cultured in increasing doses of bortezomib (Selleck Chemicals, Houston, TX), until they could be cultured in 20nM bortezomib (5TGM1 .BR). The engraftable bortezomib-resistant 5TGM1 (5TGM1.BRE) cells came from C57BL/KaLwRij mice bone marrow which were routinely cultured in 10nM bortezomib. Both Bortezomib resistant cell lines were removed from proteasome inhibitor for at least 72h before use in experiments. RNA was extracted from 1x10^7 cells per sample using the Qiagen RNeasy mini kit and RNA-seq was performed.
