Using secretion encoded single-cell sequencing (SEC-seq), which exploits hydrogel nanovials to capture individual cells and their secretions, we simultaneously measured the secretion of vascular endothelial growth factor A (VEGF-A) and the transcriptome for thousands of individual mesenchymal stromal cells (MSCs). Overall design: We combined the ability to capture cellular secretions with antibody-conjugated nanovials with a single cell sequencing workflow, which allows us to quantify VEGF-A protein secretion levels and whole transcriptome information from each single cell. We tested this SEC-seq workflow in MSCs, which can be induced to secrete more VEGF-A via hypoxic conditions.
