The design of gene detection chips heavily relies on the hybridization efficiency of oligonucleotide probes. Notably, the hybridization experiment using 3100 21bp probes targeting specific sequences only serves as an external validation set for addressing this issue. It verifies the discrepancies between traditional thermodynamic models and experimental outcomes, and supplements the gaps in existing datasets by providing large-scale, standardized hybridization data (enabled by the 21bp probe design and sandwich hybridization strategy).
