Optical Genome Mapping Raw Data

Optical genome mapping raw molecule files of the UM-SCC-47 and HeLa cell lines. Created using the Bionano Prep SP-G2 Blood & Cell Culture DNA Isolation Kit (PN 80060). Briefly, DNA was isolated from a compact cell pellet following Bionano Prep SP-G2 Frozen Cell Pellet DNA Isolation Protocol, rev C. This DNA was fluorescently labeled at the recognition site CTTAAG with the enzyme DLE-1 and counter-stained, using the Bionano Prep DLS-G2 Labeling Kit (PN 80046) following Bionano Prep DLS-G2 Protocol, rev E. Fluorescent imaging of single molecules was performed on the 2nd generation Bionano Saphyr system with instrument control software (ICS) version 5.3.23013.1, collecting approximately 300X effective coverage of the reference hg38 after automatic filtering for noise and molecule quality.
