资源简介:
Files contain molecular data associated with the project "Fate of Litter Derived VOC-C in Soil."
Project Description: To determine the influence of volatile organic compounds (VOCs) on soil, microcosms containing 13C litter types eucalyptus, stiltgrass, switchgrass, sycamore, or tulip poplar were incubated for 90 days with a common agricultural soil. Soil and litter were physically separate, but shared a headspace allowing for diffusion of VOCs from the decomposing litter to the soil. Also included was a no-litter/soil-only 'control' microcosm. There were 4 replicates per treatment (i.e. 6 treatments x 4 replicates = 24 microcosm units).
Molecular Analyses (Fierer Lab, CU-Boulder): Soil microbial community diversity composition of was assessed by sequencing 16S rRNA gene sequences (515F-806R) and ITS1 sequences (ITS1F-ITS2R). Each microcosm unit was extracted and amplified in triplicate (i.e. 24 units x 3 replicates = 72 samples). Multiplexed amplicons were sequenced on the Illuminia MiSeq instrument using 2 x 150 bp paired-end chemistry at the BioFrontiers Sequencing Facility at the University of Colorado Boulder (Boulder, CO, USA) in May, 2018. Exact sequence variants (ESVs) were inferred using the R package dada2 v1.8 (Callahan et al., 2016).
Files: Files include mapping files (mapping.txt), ESV tables (tab.txt), taxonomy files (taxonomy.txt), and sequence fastas (seqs.fa) for 16S and ITS sequencing runs.
## 2018.08_fierer_litter_voc_16S_mapping.txt ## 2018.08_fierer_litter_voc_ITS_mapping.txt## 2018.08_fierer_litter_voc_16S_tab.txt ## 2018.08_fierer_litter_voc_ITS_tab.txt ## 2018.08_fierer_litter_voc_16S_taxonomy.txt## 2018.08_fierer_litter_voc_ITS_taxonomy.txt## 2018.08_fierer_litter_voc_16S_seqs.fa## 2018.08_fierer_litter_voc_ITS_seqs.fa
