To obtain the raw STARR-seq data for identifying cis-regulatory elements, this study adopted the following experimental workflow: first, plasmid constructs containing 600-1500 bp test sequences were transfected into cotton protoplasts; then, fluorescence-activated cell sorting was used to isolate cell populations exhibiting GFP fluorescence (indicative of enhancer activity); and finally, high-throughput STARR-seq libraries were constructed using the SMART-seq2 method.
