The role of Trim58 in acute inflammation is so far unclear. The aim of this study was to determine the effects of Trim58 deficiency on acute DSS-colitis in mice. We used microarrays to assess gene expression profiles in middle colons of Trim58-/- vs. Trim58+/+ mice after inflammatory stress-mediated mucosal damage during the regeneration phase. We generated mice completely lacking the gene Trim58 (Trim58-/-). Female Trim58-/- and Trim58+/+ mice (9 weeks old, SPF) were treated with dextran sodium sulfate (DSS). In brief, colitis was induced by addition of 4.0% (wt/vol) for 7 days of DSS (molecular weight, 36,000-50,000) in drinking water and followed by normal drinking water for 5 days. On day 12, mice were sacrificed and total RNA was extracted from middle colons (n=4 mice/group). All extracted RNA samples were analyzed independently. Microarray processing was performed at an Affymetrix Service Provider and Core Facility, “KFB - Center of Excellence for Fluorescent Bioanalytics” (Regensburg, Germany).
