For the purification of B1a cells and DP cells, PECs were isolated and stained with FITC-conjugated anti-mouse CD43 (000000; BD Pharmingen, San Diego, CA), PE-conjugated anti-mouse alpha beta -TCR (000000; BD Pharmingen), PE-conjugated anti-mouse gamma delta -TCR (000000; BD Pharmingen), and APC-conjugated anti mouse CD19 (000000; BD Pharmingen). After staining, the CD43+ CD19+ TCR- B1a cells and TCR+ CD19+ DP cells were sorted to over 95% and 85% purity, respectively, with FACSAria III cell sorter (BD biosciences, San Diego, CA).
