Gene expression profiling of ovaries collected from wild type (WT) mice and progesterone receptor (PR) knock out mice

Previous studies have shown that PR is a critical regulator of ovulation. The PR-null mice (PRKO) failed to ovulate due to a failure in the rupture of the preovulatory follicles. To experimentally induce ovulation, the wild type and PR null mice (C57BL/6, 24-28 d old) were injected ip with 5 IU of PMSG and 48 h later by 5 IU of hCG. Ovaries were collected at 11 h and total RNA was analyzed using Affymetrix mouse arrays
既往研究表明,孕激素受体(Progesterone Receptor, PR)是排卵过程的关键调控因子。PR基因敲除小鼠(PRKO)因排卵前卵泡破裂失败而无法正常排卵。为通过实验手段诱导排卵,对品系为C57BL/6、日龄24~28天的野生型小鼠与PR基因敲除小鼠,先以腹腔注射方式给予5 IU孕马血清促性腺激素(Pregnant Mare Serum Gonadotropin, PMSG),48小时后再注射5 IU人绒毛膜促性腺激素(Human Chorionic Gonadotropin, hCG)。分别于11小时时采集卵巢组织,提取总RNA并利用Affymetrix小鼠基因芯片开展分析。
