In this project, we aim to gain a genome-wide understanding of the transcriptional regulatory network (TRN) in Streptomyces albidoflavus J1074. S. albidoflavus J1074 is an ideal host for heterologous expression of biosynthetic gene clusters (BGCs) from other Streptomyces species, and the TRN may play a key role in modulating its metabolic capabilities and responses to environmental stimuli. To explore the structure and function of the TRN in S. albidoflavus J1074, we have generated 91 transcriptomes across 43 unique growth conditions, including different carbon sources, salinity and pH and temperature stresses, and over time. Combining these transcriptomes with other high quality publicly available data, we have used Independent Components Analysis, a powerful unsupervised machine learning method, to generate Independently Modulated Gene Sets (iModulons), which are groups of co-expressed genes that reflect the activity of transcriptional regulators. By identifying and characterizing the iModulons in S. albidoflavus J1074, we aim to reveal the regulatory logic and dynamics of its TRN.
