M2 macrophage promoting IL-10+B cell production and contribute to alleviate asthma in mouse

B cells play a crucial regulatory role in diseases. Macrophages exist in the disease microenvironment and interact with tissue cells as well as various immune cells, but their relationship with B cells has never been reported. Using an asthma animal model and macrophage depleting agents, we found that macrophage removal significantly aggravated asthma symptoms and significantly reduced the expression of IL-10+ B cells in the asthma model. We further analyzed and found that the macrophages interacting with IL-10+ B cells in the asthma microenvironment were M2 macrophages. Moreover, through sequencing, we preliminarily found that M2 macrophages promote B cell production of IL-10 through oxidative phosphorylation pathways. Together, the results indicate that M2 macrophages act on IL-10+ B cells, thereby alleviating asthma in mouse models. Overall design: To investigate the underlying mechanism about how M2 macrophages skew B cell towards regulatory profile, we collected naïve B cells(UN), anti-CD40 stimulated B cells(B) and M2-educated B cells(M) for RNA-seq.
