iPS2-CITE-seq with arrayed polyclonal hiPSC targeting of either SMAD2 or B2M

We transfected hiPSCs WTC11 with specific shRNAs targeting SMAD2 or B2M. hiPSCs transfected were selected, cultured, and four clones were selected per targeting and expanded for follow-up validations. We cultured hiPSC and differentiated them into cardiac organoids for 7.5 days. This experiment allowed to show how the arrayed format of iPS2-seq can work and demonstrated that the clones behave in a similar way. Additionally, the CITE-seq on B2M demonstrated that CITE-seq can work in combination with iPS2-seq and potentially can help in assessing the level of KD, since mRNA expression is not always reliable, especially when expression is low.
