This dataset is derived from the doctoral research project <i>“Using Lung Organoid as a Tool to Explore Congenital Pulmonary Airway Malformations (CPAM)”</i> conducted at the University of Hong Kong. The study investigates the developmental and molecular mechanisms underlying CPAM — a common congenital lung anomaly characterized by cystic airway malformation and disrupted epithelial differentiation. Using both patient-derived and induced pluripotent stem cell (iPSC)-derived human lung organoids, the research integrates single-cell RNA sequencing, molecular assays, and functional experiments to delineate the interplay between genetic dysregulation and environmental exposure in CPAM pathogenesis.The dataset encompasses several components: (1) processed single-cell RNA sequencing expression matrices and metadata from normal, CPAM, and iPSC-derived organoids; (2) quantitative PCR, ELISA, and protein assay data validating FGF, NOTCH, PPAR, and HIF signaling alterations; and (3) experimental results from a cigarette smoke extract (CSE) exposure assay performed on organoid-based airway models, demonstrating that CSE significantly increases mucus viscosity and inflammatory cytokine secretion—particularly in diseased organoids.Collectively, these data provide an integrated molecular and functional atlas of congenital lung malformation, offering a resource for researchers interested in epithelial–mesenchymal signaling, organoid biology, and the environmental modulation of airway disease. All data have been de-identified and formatted for reuse in downstream bioinformatic and translational analyses.
