Enhanced reconstruction of Holocene bryophytes from sedimentary ancient DNA using bryophyte-targeted primers: OBITools script and reference library

This dataset contains the OBITools scripts and the FASTA file of the bryophyte reference library used to process raw sedimentary ancient DNA (sedaDNA) metabarcoding data from Lake Jøkelvatnet for the reconstruction of Holocene bryophyte diversity. The bioinformatic workflow includes demultiplexing, primer and adapter trimming, quality filtering, dereplication, denoising, chimera removal, sequence clustering, and taxonomic assignment. The reference library FASTA file comprises both a global bryophyte sequence dataset and a curated European subset, which were used for taxonomic identification during the analysis.
