Mutation produces genetic variation and is the ultimate engine of evolution. The mutational processes are shaped by both genetic backgrounds and environment. Thus, accurate quantification of mutation rates and spectrum under drugs has important implications for controlling diseases, including cancer. Here, we applied the yeast mutation accumulation paradigm to profile the genome-wide mutational footprints of two drugs across 10 distinct genetic backgrounds. Together with comprehensive time-resolved phenotyping of cell growth and viability and genome sequencing, we estimated the mutagenic effects of these drugs.
