Microarray analysis comparing the gene expression of three tissues laser-microdissected from 1-month-old lenses in mice. The original study aimed to discover additional genes that belong to a distinct subset called late fiber genes which are expressed in cells bordering the organelle-free-zone (OFZ) of the lens. Cells were harvested from three layers of the lens by laser-microdissection. RNA was isolated from each layer in order to conduct microarray analysis.
本研究采用微阵列(microarray)分析,对比了从1月龄小鼠晶状体中通过激光显微切割获取的三类组织的基因表达情况。此项初始研究的目标是发掘隶属于一类特殊亚群的新增基因,该亚群被命名为晚期纤维细胞基因(late fiber genes),其在晶状体无细胞器区(organelle-free-zone, OFZ)边缘的细胞中表达。实验中,研究人员通过激光显微切割从晶状体的三层结构中采集细胞,并从每一层分离RNA以开展微阵列分析。
