Giardia lamblia, a single-celled eukaryotic parasite, is a major cause of waterborne infection throughout the world. G. lamblia infects a wide range of mammalian hosts and is subdivided into eight genetically well-defined assemblages; named A through H. Previously, molecular studies have revealed that most of the assemblages are associated with animal infection, whereas only AI, AII, and B are responsible for human infection. However, fragmented genomes and lack of comparative analysis within and between the assemblages impede the understanding of the molecular mechanisms of host specificity, and differential diseases outcome due to giardiasis. Thus, in the current study, we are conducting comparative genomics using assemblage A and assemblage B isolates derived by axenic culture utilizing a hybrid Nanopore/Illumina sequencing approach.

溶组织阿米巴(Giardia lamblia),一种单细胞真核寄生虫,是全球范围内水传播感染的主要原因。溶组织阿米巴可感染多种哺乳动物宿主,并根据遗传学特征细分为八个明确的基因群,分别命名为A至H群。既往分子研究表明,其中大部分基因群与动物感染相关,而仅AI、AII和B群对人类感染负责。然而,由于基因群的基因组片段化以及缺乏群内群间比较分析,阻碍了对宿主特异性分子机制以及由于溶组织阿米巴病引起的不同疾病结局的理解。因此,在本研究中,我们正在采用混合Nanopore/Illumina测序方法,利用由无菌培养获得的A群和B群分离株,开展比较基因组学研究。
