RNA was isolated from two-dimensional hepatic monolayer cultures derived from liver organoids generated from 7-week-old male mice on a C57BL/6J background. Two genotypes were used to establish the cultures: wild type (Mfge8+/+) and Mfge8 knockout (Mfge8-/-) mice. Organoids were dissociated into single cells using enzyme digestion and plated on Matrigel-coated plates to generate uniform hepatic monolayers. Cultures were maintained under standard conditions, and total RNA was extracted using the RNeasy Plus Mini Kit, with RNA integrity confirmed prior to sequencing. These samples represent untreated baseline hepatic epithelial cultures and were collected to examine genotype-dependent transcriptional differences associated with the absence of MFG-E8. This BioSample corresponds to RNA prepared from hepatic monolayer cultures and contributes to a dataset investigating molecular pathways regulated by MFG-E8 in liver epithelial cells.
