Modular organization of the motor space in the mouse superior colliculus

In order to characterize the genetic diversity within the Stratum Griseum Intermediale (SGI), we screened gene expression by extracting RNA from sensory and motor layers of the Superior Colliculus (SC) at post-natal day (P)7. Sensory and motor domains were manually dissected from P7 mice in order to obtain four samples from each mouse: sensory anterior SC, sensory posterior SC, motor anterior SC and motor posterior SC. RNA was extracted from these samples RNeasy Mini kit (Qiagen, Venlo, Netherlands) and cDNA libraries were generated from total RNA using a TruSeq® Stranded Total RNA Library Prep Kit (Illumina, San Diego, CA) and sequenced on an Illumina HiSeq 4000 machine (single read, 50bp read length).

为表征中间灰质层(Stratum Griseum Intermediale, SGI)内的遗传多样性,本研究通过提取出生后第7天(post-natal day 7, P7)小鼠上丘(Superior Colliculus, SC)感觉层与运动层的RNA开展基因表达筛选。研究人员手动解剖P7小鼠的感觉与运动区域,每只小鼠可获取四份样本:感觉前区上丘、感觉后区上丘、运动前区上丘及运动后区上丘。随后使用RNeasy Mini试剂盒(凯杰公司,荷兰芬洛)从上述样本中提取总RNA,采用TruSeq® 链特异性总RNA文库制备试剂盒(Illumina, 美国加利福尼亚州圣地亚哥)以总RNA构建cDNA文库,并在Illumina HiSeq 4000测序仪上完成测序,测序方式为单端测序,读长为50bp。
