We identified adipocyte enhancer binding protein 1 (AEBP1) as a novel cancer-associated fibroblast (CAF) marker in oral squamous cell carcinoma (OSCC). To study the function of AEBP1 in CAFs, we knocked down AEBP1 in CAFs isolated from surgically resected primary OSCCs. We found that genes associated with cell cycle were significantly affected by AEBP1 knockdown in CAFs. CAFs (1^10E6 cells) were transfected with 100 pmol Silencer Select Pre-designed siRNA (AEBP1 siRNA1, s1145; AEBP1 siRNA2, s1146; Thermo Fisher Scientific) or a Silencer Select Negative Control No. 1 siRNA (Thermo Fisher Scientific) using a TransIT-X2 Dynamic Delivery System (Mirus Bio) and were incubated for 72h.
