RNA seq of femur and humerus bone tissues from Prx1cre driven Piezo1/2 mutant pups at the age of P0

The Prx1cre driven Piezo1/2 knockout mice showed bone fractures caused by reduced bone formation. In order to understand the underlying mechanism, we performed RNA seq experiments using total RNA from the bone tissue of control and Piezo1/2 mutant P0 pups. Total RNA from P0 femur and humerus bone tissue devoid of bone marrow were prepared using Rneasy Mini Kit. Three different genotypes of mice were examined: Wild type, Piezo1 CKO and Piezo1/2 DKO. 2 replicates for each group.由Prx1cre驱动的Piezo1/2基因敲除小鼠出现骨形成减少所致的骨折。为探究其潜在分子机制，本研究以对照组与Piezo1/2突变型P0仔鼠的骨组织总RNA为实验材料，开展RNA测序（RNA seq）实验。本研究采用RNeasy Mini 试剂盒，从去除骨髓的P0仔鼠股骨及肱骨骨组织中提取总RNA。本次实验共检测三类小鼠基因型：野生型（Wild type）、Piezo1条件性敲除（Piezo1 CKO）及Piezo1/2双敲除（Piezo1/2 DKO），每组设置2个生物学重复。
