Ubiquitination-directed protein degradation is important for tumor initiation and maintenance in many cancers. E3 ligases containing HECT domains are emerging as new therapeutic targets. In contrast to many other E3 ligases, the role of Huwe1 in cancer has been controversial, and its role in ovarian cancer in which Huwe1 is dysregulated remains to be clarified. Here, we showed that Huwe1 knockout inhibits the transformation of mouse ovary surface epithelium (MOSE) cells without significant effect on cell survival and apoptosis and the loss of Huwe1 after tumor formation inhibits tumor growth. In Huwe1 knockout cells, the expression of the histone H1.3 protein increased with the subsequent inhibition of H19 expression. H19 knockdown can mimic the effects of Huwe1 knockout. H1.3 silencing can partially rescue the expression of H19 and the phenotype induced by Huwe1 knockout. These data demonstrate the essential role of Huwe1 in ovarian cancer
