Gene expression profiling of mouse liver cells

We generated the individual transcriptomes of 96 liver cells using the Fluidigm C1 platform. In brief, a suspension of cells was prepared from the liver of a 14-week old B6CastF1 (C57Bl/6J mother x CAST/Ei father) female mouse and loaded onto a 10-17 m C1 Single-Cell Auto Prep IFC (Fluidigm), and cell capture was performed according to the manufacturers instructions.本研究采用Fluidigm C1平台，构建了96个肝细胞的单个转录组（transcriptome）。具体操作简述如下：从14周龄的B6CastF1雌性小鼠（由C57Bl/6J母鼠与CAST/Ei父鼠交配获得）的肝脏中制备细胞悬液，将其加载至10-17 μm的C1单细胞自动制备集成流体回路（IFC，Fluidigm），并按照制造商的操作指南完成细胞捕获。
