This study evaluates the in vivo efficiency and transcriptomic impact of the AMBER(ADAR2 Mimic C-to-U Base Editing on RNA) system, a programmable C-to-U RNA editor, for guide RNA (gRNA)-dependent editing of the STAT3 transcript in the mouse liver. We performed RNA sequencing on liver tissue from three experimental groups: a mock control (Ringer's solution), a gRNA-negative control (plasmid expressing the AMBER editor without gRNA), and a test group (plasmid expressing both the AMBER editor and a STAT3-targeting gRNA). This dataset allows for the comprehensive analysis of on-target editing efficiency, potential off-target effects, and global transcriptomic changes induced by the AMBER system in mice liver.
