SKBR3 cells were treated with DMSO (as control), lapatinib, GSK126 or lapatinib + GSK126 for 24 h to identify genes and pathways differentially upregulated/downregulated by these treatments. Overall design: SKBR3 cells were treated with DMSO or 5 µM GSK126 starting at day -6. At day -3, cells were split and continued to be treated with DMSO or GSK126. At day -1, cells were seeded at 350,000 cells per 6-cm plate in DMSO or GSK126. At day 0, cells pre-treated with DMSO were treated with DMSO or 100 nM lapatinib, while cells pre-treated with GSK126 were treated with 5 µM GSK126 or 5 µM GSK126 + 100 nM lapatinib. 24 h later (day 1), RNA was extracted using a RNeasy Plus Mini kit (Qiagen) and sequenced in an Illumina NextSeq500 run with single-end 75 bp reads.
