This study aims to dissect the genetic basis of ascorbic acid (AsA) accumulation in mango fruit by leveraging the phenotypic variation observed between the cultivars 'Jin-Hwang' (low AsA) and 'Irwin' (high AsA). The core of the investigation involves the whole-genome resequencing of these two parental cultivars . The analysis of the resulting genomic data is designed to identify genetic variants, such as single nucleotide polymorphisms (SNPs) and insertions/deletions (InDels), specifically within a previously mapped quantitative trait locus (QTL) region associated with AsA content . A primary goal is to pinpoint candidate genes within this QTL interval and to elucidate the causal mutation, such as the premature termination codon discovered in the MiGST1 gene of 'Jin-Hwang', which is predicted to disrupt gene function. The relevance of this work is substantial for molecular breeding, as establishing a direct link between specific genetic markers or haplotypes and a key fruit quality trait like AsA content paves the way for marker-assisted selection. This strategy can significantly accelerate the development of new mango varieties with enhanced nutritional value.
