In a TBX5-linked human cellular model of atrial disease, we examined the gene networks controlled by TBX5 during human atrial differentiation using single-cell RNA and ATAC sequencing. Overall design: Wildtype, TBX5 heterozygous and homozygous mutant iPSCs were differentiated to atrial cardiomyocytes. At day 20 or day 45 of differentiation, cells were harvested for single-cell RNA sequencing and analyzed using Seurat. Two replicates per genotype per timepoint were harvested.
