Conjugates formed by Jurkat cells with SEE-loaded Raji cells (as antigen presenting cells) were stained for Glu-MT (Alexa 594) shown in green and Tyr-MT (Alexa 647) shown in red. The raw images presented in the accompanying stack (left) were deconvolved using Huygens 3.0 software (Scientific Volume Imaging, Hilversum, The Netherlands).
The resulting stack of the deconvolved images is shown. These images were used to make the 3D reconstruction (Figure 2C and supplemental video).
