Affinity maturation of Boskar4 to increase its biological activity as a G-CSFR agonist. Therefore, two libraries (a saturation and a design library) were created and screened with an E. coli display system and FACS.
Conclusion:
The expected shift to higher fluorescence values over the course of the enrichment sorts (5 in total) was observed, and later a total of 16 affinity-enhanced unique variants were extracted from these FACS-enriched pools after 3 and 5 sorts.
