REDD1 is required for full amplitude activation of the glucocorticoid receptor. In keratinocytes lacking REDD1 both gene activation and repression by glucocorticoid fluocinolone acetonide was significantly blunted. HaCaT REDD1 KO (IB11 and IB12) and HaCaT Cas9 control cells (IB9 and IB10) grown in DMEM supplemented with 10% FBS, were switched to DMEM with Charcoal stripped Fetal Bovine Serum and 1 X Pen/Strep overnight, and then were treated with vehicle control (0.1% acetone, IB9 and IB11) or 1 μM of FA for 24 h (IB10 and IB12). RNA was extracted and used for Illumina gene expression array.
