Asthma is the chronic pulmonary inflammatory response that could lead to respiratory failure when allergic reactions exacerbate. The symptom severity of asthma displays an evident circadian rhythm with aggravated airway resistance in the early morning in patients. Here we investigate whether the circadian clock regulator Bmal1 in macrophages is involved in the pathogenesis of house dust mite (HDM)-induced lung allergy. We profile the transcriptome of alveolar macrophages (AMs) and found that Bmal1-deficient AMs possessed enhanced phagocytosis and were prone to decrease the allergy mediator thromboxane A2 (TXA2) and prostaglandin F2alpha (PGF2a) synthesis. The reduced TXA2 and PGF2a may lead to the downregulation of the eosinophil chemokine Ccl11 expression in epithelial cells.
