Roots from 10 independent plants subjected to 300 mg/l PS-COOH treatments and to control conditions were pooled separately to form single biological replicates for RNA-seq analysis. Transcriptomics analyses showed that antioxidant activity-related genes were significantly upregulated after PS-COOH treatments, indicating that oxidative stress was induced in cotton. Overall design: Roots cultured on 0 and 300 mg/l PS-COOHs concentrations for 9 days are used to extract RNA.The total RNA was extracted using TRIzol reagent (Invitrogen, California, USA). Then, the RNAs' purity and quantity were checked using an RNA Nano 6000 Assay Kit for the Bioanalyzer 2100 system (Agilent Technologies, California, USA). Afterwards, approximately 10 µg of total RNA was used for RNA-seq library construction.
