We describe a simple and reproducible culture system that can be used to generate erythroblasts with B-globin expression in culture systems. Upon treatment with Fe, erythroblasts showed advanced maturity with a high transcription of B-globin. Overall design: Single-cell RNA sequencing was performed using orthochromatic erythroblasts that maintained saturated status for hemoglobin synthesis before enucleation on day 33. single cell capture was performed using BD Rhapsody Express instrument. The samples were tagged using multiplexing kit, CTRL (PBMC_SampleTag05_hs_RSEC_MolsPerCell.csv) and IRON (PBMC_SampleTag06_hs_RSEC_MolsPerCell.csv).
