With this project we deposit the genome sequences of E.coli K12 MG1655 (sample 91964) and derivatives BW30270 (sample 91967), T1241 (sample 91971), U65 (sample 91973), and U210 (sample 91975). Strain U210 is a no-SCAR E.coli K12 strain suitable for gradual induction with IPTG and L-arabinose. The genotype of the resulting strain U210 is BW30270 rph+ dgcJ::IS1 ilvG+ delta-lacIZYA delta-araCBAD Pcp8-araE delta-araFGH flhDC+.E.coli K12 strain MG1655 is the reference strain with its genome sequence annotated as NC_000913.3. Strain BW30270 is a MG1655 derivative, in which a frame shift mutation in the rph gene was restored to wild-type. Strains MG1655 (CGSC#6300) and BW30270 (CGSC#7925) were obtained from the Coli Genetic Stock Center (https://cgsc.biology.yale.edu/). Sequencing revealed that BW30270 carries a dgcJ::IS1 allele. Strain T1241 was derived of BW30270 by correcting a frameshift mutation in the ilvG gene (Pannen et al. 2016, J. Biol. Chem. 291, 2357-2370). Strain U65 is a T1241 derivative with a deletion of the lac operon, and with modification of the ara regulon to enable gradual induction by IPTG and L-arabinose (Breddermann et al. 2017, Mol. Microbiol. 104, 664-676). U65 contains two FRT scar-sites. These scar-sites were removed by the noScar method (Reisch et al. 2015, Sci. Rep. 5: 15096. and Reisch et al 2017, Curr. Protoc. Mol. Biol. 117: 31.38.31-31.38.20) using a FRT-specific guide RNA.
