This analysis compared cardiomyocytes derived from induced pluripotent stem cells under biochemical and biomechanical conditions expected to induced maturation. Overall design: Induced pluripotent stem cells were differentiated into cardiomyocytes, purified by metabolic selection, then cultured as either monolayers (ML) or as 2D cardiac muscle bundles (2DMBs, also referred to as M2Ds, or 2D-microtissues) under different media conditions expected to induce maturation. All biologic replicates were derived from pooled iPSC-CMs from the same 3 batches to avoid batch-to-batch variability.
