Single-Cell Atlas of a Rat Model of Vaginal Birth Injury induced Stress Urinary Incontinence

Stress urinary incontinence (SUI) greatly affects the daily life of numerous women and is closely related to a history of vaginal delivery and aging. We used vaginal balloon dilation to simulate vaginal birth injury in young and middle-aged rats to produce a SUI animal model, and found that young rats restored urethral structure and function well, but not the middle-aged rats. To identify the characteristics of cellular and molecular changes in the urethral microenvironment during the repair process of SUI. We profiled 51,690 individual female rat urethra cells from 24 and 48 weeks old, with or without simulated vaginal birth injury.
压力性尿失禁(Stress Urinary Incontinence, SUI)严重影响广大女性的日常生活,且与阴道分娩史及衰老进程密切相关。本研究通过阴道球囊扩张术,对青年及中年大鼠模拟阴道分娩损伤以构建SUI动物模型,观察到青年大鼠可良好恢复尿道结构与功能,而中年大鼠则无法实现该修复过程。为明确压力性尿失禁修复过程中尿道微环境的细胞与分子变化特征,我们对24周龄与48周龄、分别施加或未施加模拟阴道分娩损伤的雌性大鼠的51690个单个尿道细胞进行了特征分析。
