Human NCI-H1299 non-small lung cancer cell line whole transcriptome sequencing

Leader and follower cell populations were isolated from the H1299 non-small cell lung cancer cell line using an image-guided selection technique known as Spatiotemporal Genomic and Cellular Analysis (SaGA). After embedding H1299 3-D spheroids into Matrigel and allowing invasion to proceed for 24 hours, leader cells (located at the front of invasive chains) and follower cells (located farther back in invasive chains) were highlighted via photoconversion of Dendra2 protein from green to red fluorescence. Cells were then isolated via FACS and cultured in 2-D. RNA-seq was performed in triplicate, using three separate passages of parental H1299 cells, three separately isolated populations of follower cells, and two separately isolated populations of leader cells (including two passages of one of the leader populations).
