Acute brain injury, mainly including hemorrhagic brain injury and ischemic brain injury, has been highly associated with an increased risk of neurodegeneration and cognitive decline. Although studies have partially confirmed these linkages, the subtype-specific molecular response to acute brain injury in a system-wide has not been fully elucidated. We employ multi-omics approaches including transcriptomics, proteomics, lipidomics and metabolomics of mouse models with three time points after modeling to interrogate the molecular linkages between acute brain injury and neurodegeneration. In addition, we validated these molecular linkages by: (1) public microarray datasets on the human prefrontal cortex, and (2) a total of 25 peripheral blood samples collected from healthy donors and patients with cerebral ischemia. At last, we performed animal behavior test on Nrgn silence mouse models to elucidate it is potential role on neurodegeneration surveillance.
