Expression profiling of CD24+ and CD24- population cells from myeloma cell lines. Results provide insight into the role of CD24+ cells in myeloma development. Keywords: multiple myeloma, cancer stem cell, CD24 The CD24 compartment in multiple myeloma is enriched in side population cells, however the functional heterogeneity within this subpopulation remains poorly defined. Myeloma cancer stem cell populations (CD24+) from ARP1, OCI-MY5 and OPM2 were isolated by flow cytometry using CD24 antibodies (Biolegend). The total RNA was isolated from the CSC populations using the RNeasy RNA isolation kit (Qiagen). The RNA was then converted to cDNA and they were hybridized to the Human Genome U133Plus 2.0 GeneChip microarrays(Affymetrix). The data was normalized using the MAS5 algorithm of the Affymetrix expression console version1.1 software (Affymetrix). A comparison of transcriptional profiles was then performed in CD24+ cell lines compared to the CD24- cell lines.
