Sequencing of chromatin remodelling complexes in Lung Cancer

The potential role of chromatin structure in cancer development is still a topic of hot ongoing debate. Several members of the SWI/SNF chromatin remodeling complexes have been described altered in different tumor types. In the present work, we have identified ARID2 as a new bona fide tumor suppressor gene in lung cancer. Recurrent ARID2 are associated with an absence of the protein and worse prognosis. Our results suggest that ARID2-deficiency alters the transcriptional programme and generates genetic instability which bolster the proliferative and metastatic potential of the cells both in vitro and in vivo. Moreover, we demonstrate that ARID2 deficiency significantly affects the sensitivity of the cells to anti-tumor therapies and could be used as marker to predict the response to immunotherapy. All these results support that ARID2 deficiency can be used as a prognostic factor for patient stratification and might be exploited therapeutically.
